INTRODUCTION

Methyl Ethyl Ketone has no color liquid with a sweet/sharp, acetone-like odor and fragrant. It is an extremely combustible liquid and vapor which is heavier than air and may spread long distant combustion and flashback are possible. MEK has highly volatility. It is irritating to eyes and aspiration tract of central nervous system depressant. High vapor concentrations may induce headache, dizziness, nasusea, drowsiness, confusion and incoordination. MEK also causes eye irritation and it is a possible reproductive hazard-may caused fetotoxicity based on animal data.

MEK is found in some fruits and vegetables and happens naturally in blood, urine and exhaled air as a minor product of normal metabolism.
The MEK TLV for an exposition of 8 h is 200 ppm.

 

EQUIPMENT AND INSTRUMENTAL PARAMETERS

The analysis of Methyl Ethyl Ketone requires a HPLC system including isocratic pump, autosampler and UV detector.
The use of a solvent degasser prevents air bubbles and ensure a stable baseline.

Sampler: Injection volume 50 μl, analytical run time 20 min..
HPLC pump: Flow rate 1.2 ml/min is recommended
Column temperature: Ambient (~25 °C)
UV detector: 334 nm

COLLECTION AND STORAGE OF PATIENT SAMPLES

Before and soon after work shift, collect 10 ml of urine in a suitable test tube.
The sample is stable up to 4 weeks at - 20 °C.

 

SAMPLE PREPARATION PROCEDURE

200 μl urine sample is added to vial with teflon cap

20 μl of hydrolysis reagent is added.

Incubation is carried out at 100°C for 60 min.

Vortex the mix for 10s and then Centrifuge at 4 000 rpm for 5 min

200 μl hydrolysis reagent is added.

10 μl of internal standard is added.

Incubation is carried out at 70°C for 30 min.

Samples prepared as above can be stored at ambient temperature for about 24 hours, at +2 to +8 °C up to 36 h, frozen below -18 °C up to 3 months.
